Teams played each other twice on a home and away basis. After the pool play, the top 4 teams played semi-finals and the final to find the winner.
The bottom two teams would play in a promotion/relegation series against the two top teams from the Currie Cup First Division.

The Natal Sharks won the 2010 Currie Cup.
Following the conclusion of the Currie Cup competitions a promotion/relegation series was played. The top placed First Division team played the bottom placed Premier Division team. The second placed First Division team played the 2nd to bottom placed Premier team.
The teams played home and away, with the winner determined by the competition points scored over the two games.
